在本文中,我们报道了一种用于细胞角蛋白片段抗原21-1 DNA(CYFRA21-1 DNA)的超灵敏荧光检测系统,用于肺癌的早期诊断。该方法使用以乙二胺四乙酸(EDTA)为金属配体的电子转移原子转移自由基聚合(ARGET-ATRP)。首先,通过交联剂将硫醇化肽核酸(PNA)与氨基磁珠(PuriMag G-NH2)连接,然后与CYFRA21-1 DNA(tDNA)杂交。随后,通过与tDNA的磷酸基结合,将Zr4 +引入MBs,并通过磷酸-Zr4 +-羧酸酯化学方法将ARGET-ATRP的引发剂引入。接下来,将Cu(II)Br / EDTA通过抗坏血酸(AA)还原为Cu(I)/ EDTA,以触发ARGET-ATRP,然后将大量的荧光素-o-丙烯酸酯(FA)分子从表面上接枝。 MB,可显着放大荧光信号。在最佳条件下,tDNA在0.1 fM至1 nM(R2 = 0.9988)的范围内具有很强的线性关系。检测限低至23.8 aM(〜143个分子)。基于ARGET-ATRP策略的荧光检测具有出色的灵敏度,选择性,出色的抗干扰性能和成本效益。这些结果表明该策略在生物学检测和早期临床诊断中具有相当大的潜力。
