在实验中,超顺磁性氧化铁(SPIO)纳米粒子(NPs)的聚集可大大增强磁共振成像(MRI)T2加权成像和近红外(NIR)吸收。在这项研究中,Ac-Arg-Val-Arg-Arg-Cys(StBu)-Lys-CBT探针被设计并与单分散的羧基修饰的SPIO NPs(PuriMag Core/10)结合形成SPIO @ 1NPs,并将其用于细胞内自聚集。体外实验表明,在弗林蛋白酶过表达的肿瘤细胞中,弗林蛋白酶介导的缩合反应可诱导SPIO @ 1NPs的自聚集。而且,聚集态的NPs比单分散态的NPs具有更高的MR r2值和光热转换效率。然后,体内SPIO @ 1NP自聚集可促进精确的MRI T2成像引导的光热疗法有效杀死癌细胞。我们相信,这种基于肿瘤特异性酶指导的NPs细胞内自聚集的基本技术,对于合理合成用于肿瘤诊断和治疗领域的其他无机NPs可能是有用的。
